Executive Summary : | Wound healing is a crucial process in all living animals, with four sequential phases: hemostasis, inflammatory, proliferative, and remodeling. Extracellular vesicles (EVs) play a significant role in wound response and repair, making it essential to understand their profile for developing advanced wound healing therapeutics. This project aims to accelerate the systematic assessment of EVs role in wound healing using the Caenorhabditis elegans model. Profiling EVs RNA/protein content will help understand their effective signaling during wound healing stages and correlate these with metabolic events. The project will validate and characterize specific EVs markers, adding more knowledge to the current global EVs dataset. The project will establish a C. elegans wound model and investigate in-vivo wound healing association with EVs. Extracellular vesicles will be isolated from wound response and repair stages using ultra centrifugation, and EVs will be subjected to quantitative and qualitative checks using transmission electron microscopy and nanoparticle tracking analysis. The EVs data will be screened through in silico tools, and the dataset will be submitted to the NCBI/Uniprot/Wormbase portal. The project contributes to wound healing research by profiling and validating wound healing stage-specific EVs markers, providing novel insights for wound care products and therapeutics. Results will be used for comparative wound response and repair profiles of Human EVs, potentially informing new approaches for effective therapeutic designing for wound healing. |
