Executive Summary : | Leaf rust caused by Puccinia triticina and stripe rust caused by Puccinia striiformis f.sp. tritici are two most important diseases in major wheat growing areas of India. Though there is an option to control them chemically, breeding for rust resistant varieties is most economic and environment friendly. Though several genes for both the rusts have been reported till date, most of the native and alien resistance genes have become ineffective due to continuous evolution of rust virulence(s). Identification and introduction of diverse gene(s) into cultivars provide broad spectrum resistance against a range of pathogenic races and may give sustainable genetic solution. In this context, a T. turgidum var. durum cv. Trinakria have been identified of carrying resistance to leaf and stripe rusts. Molecular mapping of leaf rust resistance gene, located leaf rust resistance gene, LrTrk on chromosome 5BS with a closest marker at 6.3 cM. Marker assisted introgression of LrTrk in susceptible wheat varieties HD2967 and HD2932 produced Near isogenic Lines (NILs) with resistance to either leaf or stripe rusts or both the rusts suggesting no linkage between them. A NIL, HD2932+LrTrk/YrTrk, with resistance to both the rusts have been used have been used for mapping of stripe rust resistance gene. The stripe rust resistance gene YrTrk mapped on chromosome 1BS with a closest marker at 16.6cM. For effective utilization of both the genes through molecular marker assisted rapid transfer with minimum background genes around the resistance locus, the development of a high-resolution molecular map is essential. The information thus generated may be utilized in identification of co-segregating markers helping in developing leaf and stripe rust resistance wheat cultivars to stabilize the production and will also help in map-based cloning of the selected gene. |
