Executive Summary :

Polyploidy has been observed to be a mechanism for damaged tissue to repair and regenerate against oxidative, metabolic stress, and necrotic signals. In zebrafish and rodents, it has been observed to help in scarless wound healing. Similar polyploidy leads to saving hepatocytes in the liver against oxidative stress and chemical toxicity and repairs the tissue. In the current study, we propose to generate polyploidy via electrofusion in dermal fibroblasts and keratinocytes. We will check their proliferation and genetic stability. Further, we will use bilayer bioprinted (one layer of fibroblast polyploids and one layer of keratinocyte polyploid) to treat chronic diabetic ulcers.
