Executive Summary :

The project aims at the molecular characterisation of TaMSL5-D gene for stress tolerance and interaction analysis in bread wheat. This gene will be cloned in the plant expression vector under CaMV35S promoter. The expression cassette will be used for Agrobacterium-mediated genetic transformation to generate an over-expressing transgenic line of bread wheat and Arabidopsis. The transgenic lines will be analysed by genomic DNA PCR for gene integration, RT PCR for gene expression, and western blotting and ELISA for protein expression analyses. Then, those lines will be analysed for various abiotic stress tolerance such as heat, drought, and salinity, and numerous morphological and biochemical characterisations. This gene will be valuable in future genetic improvement programs.
