Executive Summary :

Sirtuins are mammalian homologues of the yeast Silent Information Regulator 2 (SIR2) protein. The human genome encodes for seven sirtuins members (SIRT1-SIRT7), which differ regarding their catalytic activities, subcellular localization, protein substrates and biological functions. SIRT3 promotes mitochondrial Electron Transport Chain (ETC) activity and in turn maintains the cellular ATP levels. For this function, SIRT3 deacetylates multiple members of the ETC, which leads to an increased ETC efficiency and enhanced energy production.
